关于含有DMSO冻存液的问题

DMSO作为普通细胞冷冻保护成分常被添加到冻存液中,但同时也是对人体有害的成分,且极易被皮肤吸收。除具有细胞毒性外,如同下列论文所述,是影响细胞分化的因子之一。因此,为获得更精确的实验结果,无DMSO的细胞保存是必须的。

● 使用DMSO将人骨髓白血病细胞株 HL-60 分化为粒细胞。1

● 使用DMSO将小鼠间充质干细胞分化为心肌细胞。 2

◆应用实例1

大鼠间充质干细胞分化能力

确认使用本产品冻存的大鼠间充质干细胞在复苏后的分化能力,并与未冻存组以及10% DMSO保存的细胞进行比较。结果显示,使用本产品冻存的细胞皆维持良好的分化性能。

检测使用本产品冻存(1周)的大鼠间充质干细胞在复苏后的存活率以及 6 h后的存活率,并与10% DMSO保存的情况进行比较。本产品无论是在刚复苏(0 h),还是贴壁后(6 h)皆显示更高的存活率。

◆应用实例2

使用 CryoScarless® DMSO-Free 简单冻存后的小鼠 ES 细胞

实验方法:

将小鼠 ES 细胞(FKHR+/+)在明胶包被的96孔板上于37°C培养48 h后,进行简单地冻存。吸走培养基并用PBS清洗,然后分别添加10%DMSO/培养基,或CryoScarless® DMSO-Free,在-80°C下保存。24 h后迅速复苏,添加加热至37°C的培养基,在37°C下继续培养。6 h后使用光学显微镜观察相衬图像。

结果:

可观察到在10%DMSO/培养基中保存的小鼠ES细胞在复苏后活细胞减少(左图),而在 CryoScarless® DMSO-Free 中保存的小鼠ES细胞状态得到改善,可观察到大部分的活细胞。

讨论:

在对多个转基因克隆的培养板一起进行冻存时(简易冻存),使用CryoScarless® DMSO-Free 更加方便。利用 CryoScarless® DMSO-Free 进行冻存,存活细胞更多,更易于未分化小鼠ES细胞的传代培养(在一定细胞密度下每2天传代一次),并可顺利进行分化实验。

背景

由于再生医学不断普及,因此急需确立可安全地培养从患者获得的间充质干细胞的方法。近年来,通过牙科治疗从拔牙的牙组织(牙髓)中获得的间充质干细胞,显示比骨髓干细胞高3~4倍的高增殖性,且没有伴随染色体异常的细胞变化。而且牙髓干细胞拥有与骨髓干细胞相同的多分化性能。由于在体外培养可大量增殖,灵活利用低癌变和成瘤风险的牙髓干细胞,对于确保医疗安全性的再生医学来说是一个极具吸引力的选择。

作为再生医学中必不可少的”细胞冻存”,已经证明CryoScarless® DMSO-Free(BioVerde公司)是一款优秀的细胞冻存液。换言之,使用该细胞冻存液冻存的人牙髓干细胞拥有与未经冻存培养的牙髓干细胞相同的增殖能与多分化能力,且未发现染色体异常。

本文着眼于牙髓干细胞活跃的增殖能力,并证实了CryoScarless® DMSO-Free不会影响细胞增殖能力。

方法及结果

从20~37岁成人拔下来的智齿分离牙髓细胞并进行无血清培养,培养至传代数为1时使用CryoScarless® DMSO-Free冻存。3个月后再次培养冻存的细胞,传代数到3时进行以下的细胞评价。同时,对未经冻存的牙髓细胞也进行了相同的评价。

使用相差显微镜观察细胞形态,确认纺锤形的成纤维细胞形态没有发生变化(图-1)。根据增殖曲线分析的结果,经过12天的培养,呈现一致的细胞增殖模式,两者间在统计学上的没有显著差异(图-2)。另外,通过Bromodeoxyuridine(BrdU)进行细胞分裂能力分析的结果显示,可观察到对数生长期内大量的BrdU阳性细胞,细胞数在统计学上没有显著差异(图-3)。

结论

除了本文中报告的细胞增殖能力评价之外,使用CryoScarless® DMSO-Free冻存的牙髓干细胞不仅维持了成骨细胞、脂肪细胞、软骨细胞的多分化能力,也能维持向免疫缺陷小鼠的皮下移植并引起硬组织形成的能力。并且该产品并不影响干细胞的特性。

另外,值得注意的是,使用CryoScarless® DMSO-Free冻存的牙髓干细胞即使长期培养至传代数为10后仍然保持染色体的二倍体和正常核型,大量培养的细胞中也没有发现染色体异常。

在再生医学中必不可少的”细胞冻存”中,作为冷冻保护剂使用的Dimethyl sulfoxide(DMSO)可能会有细胞毒性和引起意外的细胞分化,因此,我们十分期待能够确立一个像该产品一样的无DMSO的细胞冻存体系。

本报道中使用易于获得的牙髓来源间充质干细胞,研究了CryoScarless® DMSO-Free对干细胞特性的影响,并确认了使用该产品冻存的细胞与未经冻存的牙髓干细胞拥有相同的细胞特性。因此,CryoScarless® DMSO-Free在间充质干细胞基础研究以及临床前研究,甚至是将来的再生医学领域中,有望成为一款可以提供安全且高预测性数据和治疗成果的细胞冻存液。

◆操作方法概要

1. 将培养细胞放入离心管,离心并去掉上清。

2. 按照5×105~5×106 cells/mL添加本产品,并制成细胞悬液,每个冻存管分装1 mL。

3. 放入-80℃超低温冰箱中冻存。

4. 将在上一步中保存的细胞放入37°C恒温水浴槽中,快速晃动使细胞解冻,融化后放入适当的培养基(10 mL)中并离心清洗。

※长期保存请置于液氮罐中保存。

使用说明:
1、溶液的准备
偶联液:0.1M Na2CO3, pH8. 5-10.0
封闭液: 1M乙醇胺, pH8.0
清洗液1: 0.1M 乙酸~乙酸钠,0.5M NaCI, pH4.0
清洗液2: 0.1M Tris-HCl, 0.5M NaCI, pH8.0
保护液:含20%乙醇的1XPBS
2、样品准备
样品用偶联液溶解,浓度约5-10mg/ml。
3、样品偶联
1)取适量的环氧活化琼脂糖凝胶6B,去除保护液,离子水清洗三次,用偶联液清
洗一次。
2)将溶解好的样品中溶解后转入清洗好的环氧活化琼脂糖凝胶6B中填料:样品.
溶液体积比约为1:1。
3) 25- 40°C摇床中振荡混合反应24h。注:确保树脂悬浮起来,否则会大大影响偶联效率。
4)反应完后收集偶联样品,以便检测偶联效率。偶联液清洗一遍。
5)加入等体积的封闭液,37 度振荡混合反应1h。
6)将上述反应体系取出,流干其中的溶液,用3倍柱体积的去离子水清洗树脂,清洗液1、去离子水、清洗液2和去离子水
重复冲洗2次,然后保存在等体积的保护液中,于29C-89C保存。
注意事项:
1.不同配基的偶联方法与上面类似,但是反应的条件例如:配基加入量,反应时间,反应温度应根据配基的不同进行适当的
调整
2.可用碳酸盐、硼酸盐和磷酸盐缓冲液做为偶联缓冲液,但绝对不能使用含有氨基的Tris、甘氨酸的缓冲液。

下面简要介绍环氧活化琼脂糖微球偶联含氨基配基的使用过程

环氧活化琼脂糖微球FF 是将环氧基团键合在琼脂糖微球 6FF 上冻干形成的一种活性中间体,可直接偶联含氨基、巯基和羟基的配基制备分离介质,用于生物大分子的纯化分离。

  • 干粉的溶胀(非干粉形态忽略此步骤)

称取一定量的环氧活化高流速琼脂糖微球冻干粉,在蒸馏水中悬浮溶胀(1g 干粉约溶胀为 3~4ml 凝胶),放置大约 1 小时后抽滤,并用约 200ml(PH 7)去离子水洗涤干净其中的添加剂,并在漏斗中抽干。

  • 配基的偶联

一般配基偶联的过程如下:

  • 偶联溶剂:可用去离子水、碳酸盐缓冲液和磷酸盐缓冲液。不能用含氨基和其他含亲核试剂的溶液。有时需要有机溶剂溶解配基,如用 50%二甲基甲酰胺和二氧六环等。
  • 取一定量的配基溶解到偶联用的缓冲溶液中,使其完全溶解,最低浓度为200μmol

配基/ml 凝胶。凝胶与缓冲溶液以 1:0.5 到 1:1 的比例混合悬浮反应。

  • 将溶胀的凝胶悬浮到含有配基的缓冲溶液中,在20℃~45℃的水浴中摇荡 16 小时。也可以用其他搅拌方法使配基偶联,请勿用磁力搅拌。
  • 反应完全后,再洗涤掉缓冲液中的多余的配基。
  • 残余活性基团的消除,将反应完洗涤过的凝胶悬浮到 1mol/L乙醇胺(PH 8)的溶液中,在 40℃~50℃最少反应 4 小时或者室温反应过夜。
  • 在反应完后,用含有 5mol/LNaCl 的 0.1mol/L 醋酸缓冲液(PH 4.0)和含有 0.5mol/LNaCl 的偶联缓冲液(PH 8.3)交替洗涤,至少重复以上循环洗涤 3 次。
  • 装柱

⑴让所有的材料和试剂达到室温。配制初始缓冲液(平衡液)和洗脱缓冲液。

⑵根据柱子大小取所需量的凝胶,清洗掉溶液,抽干,用初始缓冲液(按凝胶:缓冲液

=3:1  的比例)配成匀浆。匀浆作脱气处理。

⑶将柱内及柱子底端用水或缓冲液润湿并保持一小段液位(液面略高于滤膜),务必使底端无气泡。

⑷用玻璃棒引导匀浆沿着柱内壁一次性倒入柱内,注意勿使产生气泡。打开柱子出液口,使凝胶在柱内自由沉降,连结好柱子顶端柱头。

⑸打开蠕动泵,让缓冲液用使用时流速的 1.33 倍的流速流过,使柱床稳定。用 2~3 倍柱体积的缓冲液平衡柱子。

  • 平衡

让平衡缓冲液以一定流速流过柱子,到流出液电导和 pH 不变。平衡缓冲液一般是根据需要纯化的蛋白来决定用哪种缓冲液。

  • 上样

⑴样品用平衡液配制,浑浊的样品要离心和过滤后上样。

⑵一般情况是让目标产品结合在柱子上,用平衡液洗去杂质,再选择一种洗脱液洗下目标产品。

⑶介质对样品组分吸附的程度取决于介质配基的亲和作用、样品的亲和性质、流动相的组成和温度。

(4)再用缓冲液平衡到流出液电导和 pH 不变。

  • 洗脱

洗脱液也要根据需要纯化的蛋白决定。

  • 再生

对于使用之后的凝胶,以 2~3 倍柱体积的 0.1mol/L Tris·HCl+0.5mol/L NaC(l PH=8.5)

和 0.1mol/L NaAc+0.5mol/L NaCl(PH=4.5)依次反复冲洗 3~4 次,进行再生。

  • 在位清洗

根据偶联的配基选择在位清洗的方法,以洗去柱子污染物同时不伤害柱子为原则。对于一般的污染,用 0.1mol/L NaOH 进行较长时间的清洗,对配基基本没有的影响;当严重污染后,可用 0.5mol/L NaOH  进行短时间的清洗;对于介质中含有其他试剂的,可用含有 8 mol/L尿素和 6 mol/L  的盐酸胍的溶液进行在位清洗。若有失活蛋白或脂类物质洗不干净,可用非离子表面活性剂如 0.1% Triton 洗涤。

清洗完毕后,用至少 3 倍缓冲液平衡柱子。

  • 注意

在装柱、使用和保存柱子的时候,要避免柱子流干,气泡进入。

简介
DEAE-纤维素,它采用平均粒径为50μm的颗粒型亲水高分子聚合物,表面又用大
分子糖链接枝,使它有更高的比表面积和更好的生物兼容性,它在高流水下保持
更高载量,同时又具有更好的分辨率。由于比表面积大,平衡和洗脱的时间也更.
短。它经过接枝即使是纯化病毒,质粒等超大分子的物质,载量基本保持不变。

关于DEAE52纤维素的处理只有简单的几步,如下:

1.先将干粉的纤维素浸泡在蒸馏水中,一段时间大约3小时左右,我偏爱这个时间,去除杂质,最好抽干一下;
2.再用0.5mol/l的HCl溶液浸泡2小时,用去离子水洗净至PH中性,并抽干;
3.将抽干的纤维素再浸泡在0.5mol/l的NaOH溶液中2小时,用去离子水洗至中性,抽干。即可用了
对于用过的纤维素,可以重复利用多次,但需要经过再生处理后才可以使用。再生处理可先用高浓度NaCl (1-2 mol/L)过柱冲洗柱床,以除去DEAE-52阴离子交换纤维所吸附的成份。然后,再用0.5 mol/L的HCl和NaOH处理,处理方法与预处理完全相同。

DEAE-纤维素的处理及装柱
(1)处理
本实验采用的是DEAE-纤维素DE 52 是弱酸型阴离子交换剂,具体处理方法为:先将DE52阴离子交换剂干粉浸泡于蒸馏水中,去除杂质;再在0.5N的HCL溶液中浸泡1-2h,再用无离子水或蒸馏水洗至pH值中性或PH4以上,并将其在抽滤漏斗中抽干;将抽干的离子交换剂浸泡在0.5N的NaOH溶液中1-2h,再用无离子水或蒸馏水将其洗至中性。
(2)装柱
将层析柱清洗干净垂直固定到层析架上,加1/3体积的无离子水,打开下出液口,水流畅通,即刻将小烧杯中装有适宜浓度的柱材,轻轻倒入层析柱中,凝胶自然慢慢沉降再层析柱底部,凝胶沉积直到离层析柱上端1.5-2cm处,停止装柱。层析柱上端进液口连接恒流泵,下出口连接蛋白质监测仪,待层析柱的平衡。
(3)平衡

在柱层析上样前必须对层析柱进行平衡,所谓平衡就是将层析柱中的溶液用层析过程的缓冲液(洗脱液)置换出来,使层析柱中的缓冲系统与柱层析过程中的系统一致。其方法是:利用层析柱上端的恒流泵将平衡缓冲液泵入到层析柱内,打开层析柱下端的出口,平衡液流速在0.5-1ml/min,当下出口流出液的PH值与平衡缓冲液的PH值一致时,层析柱达到了平衡。在本实验中,用0.002M Tris-HCL缓冲液PH7.4(内含0.0001M EDTA),预先将DE-52柱进行平衡。

DEAE—纤维素的活化

称取IgDEAE32或52,放入5ml量筒中,加蒸馏水浸泡过夜,观察溶胀后DEAE的体积。根据所需层析柱的柱床体积计算所需DEAE的用量,称取所需DEAE用蒸馏水浸泡过夜,其间换几次水,每次除去细小颗粒。抽干,改用0.5ml/LNaOH溶液浸泡1h以上,抽干(可用布氏漏斗),用无离子水漂洗,使pH至8左右(用pH试纸检查)。再改用0.5ml/LHCl溶液浸泡1h以上,去酸溶液,用无离子水洗至pH6左右。本实验中在用前应以0.0175mol/L,pH6.7磷酸盐缓冲液,浸泡平衡后使用。

DEAE—纤维素的再生
用过的离子交换剂可以反复使用,使其恢复原状的方法俗称“再生”。再生并非每次用酸、碱反复处理,通常只要“转型”处理即可。所谓转型就是使交换剂带上所希望的某种离子,如希望阳离子交换剂带上NH+4则可用NH4OH浸泡,如希望阴离子交换剂带上Cl—则用NaCl溶液处理。本实验中DEAE—纤维素在酸碱处理后,用0.0175mol/L,pH6.7磷酸盐缓冲溶液浸泡即可转型,以HPO4取代DEAE中的OH—。一般由于DEAE—纤维素使用后因带有大量杂蛋白,所以再生时,先用0.5ml/L NaOH浸洗,用去离子水洗至pH8左右,再转型,即可再使用。

三、应用的注意事项:
1)色谱柱装填
1、所需要用到的材料的温度要与色谱操作的温度一样,液体最好做脱气处理。
填料可直接称量需要的量用缓冲液溶涨一小时装柱即可.
2、在柱子下端加入20%乙醇,以除去柱子中的空气,关闭柱子出口,在柱
内保留少量的20%乙醇。20%乙醇容易产生气泡,可以在里面加1%吐温避免气泡产生。也可以换成纯水装柱子,但是需要把填料中的20%乙醇也换成纯水,具体
的方法取需要体积的填料在抽滤漏斗上进行,也可以小心倾去填料上的20%乙
醇,再换成5倍体积的纯水,反复沉淀去上清,5次左右就可以用于装柱子。
3、此填料颗粒比较细,所以一定 要注意柱子要选择合适的筛网,不能漏,
也可以取点填料加到筛网.上试试,如果没有问题再将填料连续倒入柱子时,要用
玻璃棒的紧靠柱子内壁引流,以减少气泡的产生,让填料先自然沉降到填料体积
不再变化,而填料和上面的液体很好分层,上层溶液完全澄清,就可以开泵用适
当的流速压柱子,填料体积不再变化后,再把转换头紧顶在填料上就可以平衡柱
子使用。使用的流速要小于装柱子的流速。
4、在装柱子前,填料从冰箱中取出至少要室温放置2-3个小时,这样避免
装柱子时由于温度变化而使柱子中产生气泡。
2)蛋白的结合
样品的盐浓度和pH要尽量和平衡柱子的缓冲液-致,盐浓度过高或者pH带
低也许挂不上,所以要根据自己的样品做适当调整。
3)蛋白的洗脱
这个填料如果采用线性梯度洗脱,柱子的直径和高度比最好是大于10,数值
越大越有利于分离,而且样品最好别上太多,可以按约10mg/ml.上样,如果采用
阶段洗脱的方法,装短粗柱子就可以,上样量也没有限制。阶段洗脱容易放大,
重复性好,如果洗脱条件好完全可以得到和线性梯度一样或者更好。采用什么方
法完全根据自己需要。
四、再生清洗
1、每次用完最好用含2M NaCl洗5个床体积,再用水洗5个柱床体积,然后用
20%乙醇保存,使用3-5次后在水洗之后再用70%的乙醇或30%异丙醇都含1%吐
温洗5个柱床体积,最后20%的乙醇流洗5个柱床体积。
2、有机溶剂和水混合很容易产生气泡,为了避免这样情况,可以把配好的有
机溶剂在室温放置过夜,再使用,这样可以避免气泡进柱子而导致柱子不能正常
使用。
五、保存
在20%乙醇中,4’C下长期保存。

包被载玻片所用的多聚赖氨酸的分子量在150000-300000之间。首先配制0.1%的多聚赖氨酸水溶液,贮存于-20℃。包被时,取出溶液由载玻片的一段滴上,使其慢慢印迹,直至完全覆盖玻片。多聚L赖氨酸包被的膜一定要确保很薄,经过风干即可使用。在一般免疫组化实验中,多聚L赖氨酸不影响实验的结果。但是在免疫胶体金-银染的实验中,一定要谨慎,可能会引起沉淀反应。

These Superfrost Plus(+) glass slides are made by a new process which offers the following features

  • They place a permanent positive charge in the microscope slide
  • They electrostatically attract frozen tissue sections and cytology preparations, binding them to the slide
  • They form a bridge so that covalent bonds develop between formalin fixed sections and the glass
  • Tissue sections and cytological preparations adhere better to the Plus glass slides without the need for special adhesives or protein coatings

Benefits

  • Plus glass puts an end to background staining
  • It eliminates blue or red background staining which is often encountered in hematoxylin and eosin staining of tissue sections mounted on albuminized glass
  • No brown background staining occurs when immuno-peroxidase or in situ DNA procedures are performed on our Plus glass. A higher degree of enzyme digestion can be performed on plus glass without the loss of tissue sections
  • Cytological preparations placed on Plus glass resist cell loss during enzyme digestion, DNA denaturation and RNA hybridization
  • Plus glass improves tissue adherence in microwave staining
  • Chromic acid washes are redundant for molecular hybridization because the slides are RNAse-free
  • Plus slides are Color coded to help keep track of specimens during processing as well as filing

  • Soften point is about 1250°C (2282°F)

Permanox™ is a strong, biologically inert material resistant to non-chlorinated hydrocarbons, acetone, and other EM reagents. Permanox™slides allow you to do cell fixation and staining on the slide; they can be cut with scissors. Permanox™ has not been shown to be autofluorescent. All of the microscope slides are 1″ x 3″.

NOTE: Long exposure to solvents will cause the slide to warp. Xylene and Toluene based mounting media should be avoided. Glycerol, gelatin, and other aqueous based media are recommended.

These TOMO hydrophilic adhesion slides offer superior tissue adhesion, staining performance and slide surface fluidics. Proprietary slide coating ensures consistent performance even after exposure to heat stress and humid environmental conditions. Optimized for use with thermal printers.

These slides are recommended for use on Ventana® Benchmark staining platform and automated IHC platforms.

These slides are exceptionally flat, with low fluorescence glass, and finished with 90° and clipped corners.

Fluorescence Enhancing Slides and Versatile, (bio)-functionalized coatings for Glass, Metal Oxides and ITO

Fluorescence systems (e.g. microscopes) are today widely used in the life science research. For many applications an increased sensitivity compared to existing glass slides is required. The BrightSlide™ D technology uses a proprietary dielectric coating to significantly enhance the signal intensity of fluorescence molecules from the surface. It can be read out with all systems that illuminate and detect from the top. The technology makes use of purely optical principles and substitutes conventional glass substrates.

The BrightSlide™ D (Dielectric) comes standard as a bare SiO2 Surface.

Made from Corning® 7740 borosilicate, low expansion, type I glass.

Micro glass slides are precision rectangular glass capillary tubes for use in microscopic viewing of liquid samples. Capillary action instantly collects the sample and the thin glass walls minimize evaporation. The design of these cells eliminates the needs for cover glass and the hanging drop method. These micro glass slides can withstand heat up to 500°C. Samples can be retained for future reference by sealing or cementing open ends. Micro glass slide are packaged in plastic vials.

PTFE Printed Slides

PTFE Printed Slides, are extremely hydrophobic, acetone proof, chemical resistant and autoclavable. They are perfect for controlling cross-contamination in staining, immunolabeling and immunofluorescence tests. All printed slides are precleaned. The well surface is 100% wettable, which enhances cell attachment and increases its spreading capabilities within the well. These slides minimize the use of expensive purified antigens and antibodies. The slides measure 3 x 1″ (76 x 26 x 1 mm).

EMS Printed Slides are manufactured to order offering maximum shelf life. The shelf life of PTFE Printed Slides range from 18 months to two years. However, it is recommended that all glass slides (Printed or un-printed) be used in a timely manner to avoid glass “weathering”, (slides becoming cloudy or hazy even with proper storage. All of our Printed Slides are autoclavable and have a frosted end. They are intended for one time use only.

Lovins Micro-Slide Field Finder

Marking a microscope slide to identify a particular field of interest for future reference has always been a difficult problem for many microscopists.

The Field Finder has a precision rectangular-coordinate grid pattern, which is accurately positioned on a special microscope slide. The slide has been carefully edge ground to provide straight edges and square corners to ensure repeatable results.

Lovins Micros-Slide Field Finder helps you to index points of interest on a microscope slide the same way you find them on a road map. A standard 75 x 25 mm glass slide has a photo etched grid pattern of 1 mm squares, subdivided into 0.1 mm intervals. Each square is marked with a letter and a number. You center the detail in the field of the microscope, replace the specimen slide with the field finder, and read the coordinates. Note reading on slide, and you can find that detail immediately by reversing procedure.

The pattern is imaged in vacuum deposited chrome for durability.

Retriever Tissue Slides are high adhesive glass microscope slides for paraffin-embedded tissue section, including tissue arrays. The slides were designed and optimized for the EMS Antigen Retriever (62700 series) to preserve the section attachment and tissue morphology during heat-induced epitope recovery in Retriever in pH 6 (Low), pH 9 (High), or Universal Retriever buffer.

Slides can also be successfully used in any other epitope-recovery technology (autoclave, microwave, proteolitic) and show superior performance over positively charged or polylysine-coated slides, especially in high pH buffers and recovery buffers containing EDTA.

These slides are ideal for both IHC and immunofluorescent applications, as well as for all antigen recovery systems.

Each of the slides have a custom imprinted frosted colored end, each color representing a different glass surface.
