作用机制:
PPM(Plant Preservative Mixture,植物组培抗菌剂)是用于植物细胞培养中的一种强效且广谱的生物杀灭剂,由多种抗菌剂组合而成的混合物,能杀死植物组培培养基和污染组织内的细菌和真菌,作用机制在于能够穿透细菌或真菌细胞壁,抑制柠檬酸循环和电子传递链等中心代谢循环中关键酶的生物活性,以及抑制培养基内单糖和氨基酸往细菌或者真菌细胞的运输。取决于PPM剂量和污染程度,PPM可用作植物培养基内的生物杀灭剂组分。PPM还可用作生物静力化合物起到预防效果。除此之外,PPM能有效抑制植物培养液体或半固体培养中通过空气、水、人传播的微生物污染以及内源性污染,但不会影响体外种子发芽、愈伤组织生长以及根茎再生。
适用植物类型:
PPM有效作用于大多数种子裸露植物(被子植物和裸子植物),但不适合用于蕨类,藓类,藻类和水生植物。【PPM是一种非常有效的抗微生物剂以预防和去除组培污染,然而不能替代实验室的无菌处理试剂,建议使用合适的空气无菌处理系统。】

使用特点:1)PPM可在灭菌前加入培养基或者分液前直接加入灭菌培养基;
2)PPM可在压力15 psi(1.05 kg/cm2),121°C灭菌20min;
3)PPM需要灭菌后(post-autoclave)后加入含蛋白质的培养基内,之后将混合好的培养基倒入培养皿;
4)工作浓度的PPM(经培养基稀释)可室温稳定存放高达1个月;
